Eli Lilly's Retatrutide Shows Significant Weight Loss in Phase 3 Trials

Eli Lilly and Company has reported successful results from two pivotal Phase 3 trials, TRIUMPH-2 and TRIUMPH-3, demonstrating that its investigational triple hormone receptor agonist, retatrutide, achieves substantial weight reduction in adults with obesity, type 2 diabetes, and established cardiovascular disease.

In the TRIUMPH-2 trial, which focused on participants with obesity and type 2 diabetes, those treated with retatrutide experienced significant weight loss at 80 weeks, with the highest dose of 12 mg resulting in an average reduction of 49.6 lbs, or 20.8% of body weight. The drug also improved glycemic control, showing A1C reductions of up to 1.6%.

The TRIUMPH-3 study, targeting patients with severe obesity and cardiovascular disease, yielded similarly robust findings. Participants reached weight loss averages of up to 55.8 lbs, or 22.6%, over the same 80-week period. Beyond weight management, the medication showed potential in reducing cardiovascular risk factors, including notable drops in triglycerides, non-HDL cholesterol, and systolic blood pressure.

While the trials confirmed the drug's efficacy, they also tracked common adverse events such as nausea, diarrhea, and vomiting, which were generally categorized as mild to moderate. Eli Lilly plans to leverage this clinical data package to submit a Biologics License Application to the FDA in the first quarter of 2027. If approved, the company positions retatrutide as a key future tool for managing complex cardiometabolic health conditions.
