Nuravax Reports Positive Immunogenicity for Alzheimer’s Vaccine

Every participant in a Phase 1 clinical trial for the experimental AV-1959R vaccine successfully developed anti-amyloid-beta antibodies. The study, published in Molecular Neurodegeneration, suggests that the MultiTEP-based vaccine can bypass immune tolerance, marking a potential shift in how researchers approach preventive treatments for Alzheimer’s disease.

Nuravax Reports Positive Immunogenicity for Alzheimer’s Vaccine

The trial enrolled 16 cognitively unimpaired individuals, with 12 receiving the investigational vaccine and four receiving a placebo. While the study was not designed to measure clinical efficacy, the immunogenicity results were robust: vaccine recipients showed geometric mean titers of 60,240 and 69,530 depending on their dosage. In contrast, placebo recipients maintained baseline levels of 156.

Exploratory analysis indicated that the generated antibodies preferentially bound to aggregated amyloid-beta species, including fibrils and protofibrils, rather than monomeric forms. Serum samples from the vaccinated group also demonstrated an ability to bind amyloid plaques found in cortical tissue from Alzheimer’s patients. According to Dr. Michael Agadjanyan, founder of Nuravax, the platform successfully overcomes the body's natural tendency to tolerate self-proteins like amyloid-beta.

Looking ahead, the team is preparing a Phase 2 trial to test the vaccine in individuals with biomarker-confirmed amyloid pathology. This upcoming stage will expand safety monitoring to include potential complications such as ARIA-H and ARIA-E, while utilizing brain and blood biomarkers to assess surrogate efficacy. Lon S. Schneider of the Keck School of Medicine of USC emphasized that while these initial results are promising, the next phase remains essential to establish whether the immune response translates into clinical benefit.
