Carnyx Therapeutics Taps New CMO as Sleep Drug Shows Promise in Rats

Aged rats treated with the oral candidate CNYX-005 gained up to two hours of total sleep time in recent preclinical trials, prompting Indianapolis-based Carnyx Therapeutics to appoint veteran drug developer Chris Brearley, M.D., as Chief Medical Officer to lead the compound toward human clinical evaluation.

The study utilized continuous electroencephalography to track sleep architecture, revealing that the extra rest was driven by an increase in non-rapid eye movement (NREM) sleep. CEO Prof. Patrick Gunning emphasized that the focus is on restorative physiology rather than simple sedation, noting that the observed NREM boost is critical for hormone secretion and tissue repair—processes that typically decline with age.

Dr. Brearley joins the company with over three decades of experience at pharmaceutical heavyweights including Pfizer and Amgen. His arrival signals a shift toward formal clinical development for the candidate, which originated from research into peptide biology and healthy aging. While the FDA has recently scrutinized the use of similar peptides in pharmaceutical compounding, Carnyx aims to distinguish its program through systematic pharmacology and rigorous safety profiling. Beyond the sleep candidate, the firm continues to advance CNYX-001, an experimental therapy for retinitis pigmentosa.
